Characterizing the role of fibroblast migration in an in vitro assay of wound contraction

Abstract

Cells in Fibroblast-Populated Microsphere (FPM) Wound assay, in contrast to the FPM Traction assay, can exhibit circumferential orientation due to a contact guidance response: the FPM compaction is inhomogeneous due to the imposed non-uniform initial cell distribution, inducing circumferential orientation of the fibrin fibrils. The assay, in this manner, is in fact a model system for studying a basic morphogenetic process. Accordingly, the dependence of FPM compaction and cell distribution on initial cell concentration and FPM dimensions are thus experimentally described and discussed in the context of an anisotropic biphasic model for cell-matrix mechanical interactions.
